Trong nghiên cứu này, letrozole (quy mô 100g/mẻ) được tổng hợp thành công từ phản ứng giữa 4-[1-(1,2,4-triazolyl) methyl] benzonitrile (TMB) và p-fluorobenzonitrile với sự có mặt của tác nhân deproton hóa potassium tert-butoxide (t-BuOK) trong dung môi tetrahydrofuran (THF).
