4705Antifungal activity of oxidative damaging agents in combination with metal cations against Trichophyton rubrum isolated from a dermatological patient

Trichophyton rubrum is a causative agent responsible for human chronic dermatophytosis worlwide. It is a common causative agent of the dermatophyte infections and resistant to many antifungal drugs. Oxidative damaging agents belong to a strong antimicrobial group and have been intensively investigated. The synergistic oxidative damage can occur if the agents react with metal cations. Therefore, in this study the antifungal activity of formulas containing the two oxidative damaging agents, including hydrogen peroxide (H2O2) and 8-hydroxyquinoline (8HQ), in combination with metal cations has been determined to examine the synergistic damage in T. rubrum isolated from the skin of a dermatological patient. The isolate was identified by morphology observations, biochemical tests and sequence analysis of ITS1-4 gene, in comparison with the similar ITS nucleotide sequences of others.

4707Anti-inflammatory effects of the extracts from Ardisia silvestris Pitard leaves in experiment

This research aimed to explore the anti-inflammatory effects of n-hexane, ethyl acetate (EtOAc), and n-butanol extracts from the leaves of Ardisia silvestris Pitard, both in vitro and in vivo, specifically focusing on the EtOAc extract. The in vitro assessment of anti-inflammatory activity involved measuring the inhibition of nitric oxide (NO) production in LPS-stimulated RAW 264.7 macrophage cells. The findings revealed that the EtOAc extract exhibited the highest potency, with an IC50 value of 52.10 ± 2.07 µg/mL, effectively inhibiting NO production without causing cellular damage. In vivo, the EtOAc extract’s anti-inflammatory properties were evaluated using carrageenan-induced paw edema in rats and the cotton pellet-induced granuloma test in mice. At both tested dosages, the EtOAc extract significantly reduced paw edema induced by carrageenan in rats. Additionally, it effectively inhibited exudate and granuloma formation following cotton pellet implantation at both doses, indicating its anti-inflammatory activity during the subacute phase of inflammation. Overall, these results strongly suggest that the ethyl acetate extract of Ardisia silvestris Pitard possesses significant anti-inflammatory properties and may be beneficial in traditionally treating various inflammatory conditions.

4708Antioxidant and hepatoprotective effects of hypericum sampsonii

This study investigates the antioxidant activity and potential hepatoprotective of Hypericum sampsonii extract (BLD1 extract) in vitro and in vivo. The in vitro experiment was carried out with DPPH (1, 1-diphenyl-2-picrylhydrazyl) radical scavenging assay to assess the antioxidant properties of BLD1 extract. In the in vivo experiment, the hepatoprotective activity of BLD1 extract was evaluated on an acetaminophen-induced hepatotoxicity mouse model. The in vitro assay demonstrated that the extract exhibited antioxidant activity in a dose-dependent manner. In the in vivo experiments, BLD1 extract at both doses (3.6 g/kg b.w day and 10.8 g/kg b.w day) reflected the decrease in serum ALT, AST and MDA level. Hepatic histology was also investigated post-mortem to assess the degree of liver injury. The overall findings of this study demonstrate the potential hepatoprotective effect of Hypericum sampsonii in acetaminophen-induced oxidative stress and liver toxicity in mice. This study has established the efficacy of Hypericum sampsonii, which should warrant further clinical trials and applications.
